Coche Island was discovered by Christopher Columbus in 1498 and originally inhabited by the Waika Rio indigenous people.

Early 16th-century settlement attempts were influenced by refugees from Nueva Cádiz, a city destroyed by a tropical storm on nearby Cubagua Island.

However, these early settlers abandoned Coche by 1574.

The island saw successful permanent settlement only in the 19th century and has been continuously inhabited since.

It forms the municipality of Villalba, with San Pedro de Coche as its capital.
